技術說明

本發明揭露一種單步驟純化重組胃幽門螺旋桿菌嗜中性白血球激活蛋白(HP-NAP)的方法，其步驟包括：提供一含有HP-NAP之待純化樣本，其中該待純化樣本係取自一可表現重組HP-NAP之蛋白質表現系統；將該含有重組HP-NAP之待純化樣本以一二乙氨乙基(DEAE)離子交換樹脂，在一預定pH值區間進行純化；以及收集經由該上述步驟後之一流出液(flowthrough fraction)及清洗液(wash fraction)，其中該流出液及清洗液包含一預定純度之重組HP-NAP。本發明利用高親和性的DEAE Sephadex離子交換樹脂以單一步驟將水溶性蛋白的重組HP-NAP純化出來，藉此從枯草桿菌表現系統中純化出低內毒素殘留的重組HP-NAP，且純化出來的重組HP-NAP具有典型的α-helix二級結構且可活化嗜中性白血球以產生活性氧。 The present invention provides an one-step purification method for recombinant Helicobacter pylori neutrophil-activating protein (HP-NAP). The method comprises the steps of: providing a sample containing recombinant HP-NAP, wherein the sample is obtained from a protein expression system that can express recombinant HP-NAP; purifying the sample by a Diethylaminoethyl cellulose (DEAE) resin between a predetermined pH range; and collecting a flowthrough fraction and a wash fraction obtained through above step, wherein the flowthrough fraction and the wash fraction contain the recombinant HP-NAP having a predetermined purity. Purified recombinant HP-NAP has a common α-helix structure and can stimulate neutrophils to produce reactive oxygen species (ROS).
