技術說明

本研究以本實驗室多年研發多效型桿狀病毒表現載體之平台進行含有P35與P40之介白素12 (interlukin 12)的生產。我們同時引進雙tag的技術以利介白素12 之純化，避開P40 homodimer為介白素12 抑制劑之瓶頸。由3*106 High 5細胞可純化得到85 ug的介白素12 ，相當於1公升可以純化得到3.4毫克介白素12 ，而且我們也證實此桿狀病毒表現系統生產的介白素12可以誘發Jurkat cell分泌干擾素r (interferon r)，並證實其具有活性。因此此技術可用於生產具商業價值的介白素12。 We have developed muti-cistronic expression system for producing interlukin 12 containing P35 and p40 in our lab for years. In the mean time, we introduced dual-tag technique to facilitate the purification of interlukin12 to avoid P40 homodimer as the bottleneck. 85 ug of IL 12 can be purified from 3 * 10 6 High 5 cells , the equivalent of 3.4 mg of purified interleukin 12 from 1 liter medium. Moreover, we confirmed the IL12 produced by baculovirus expression system can induce interferon r secreted from Jurkat cells . Therefore, this techniques we developed can produce ecnomic valuable IL12.
