光子晶體和磁珠都為常用於增強螢光與降低偵測極限的工具,而本申請案為第一個結合光子晶體與磁珠載體的螢光免疫檢測方法與裝置,透過光子晶體的導模共振效應,搭配磁珠的高表面與體積比(surface to volume ratio)特性,再加入聚集磁珠的技巧,本申請案可達到(1)增強螢光訊號達數千倍以上,(2)偵測極限可大幅降低,如fg/ml等級,提供現有磁珠式螢光免疫分析法無法達到的偵測能力。 Photonic crystal and magnetic beads are often used to enhance fluorescence signal to reduce limit of detection, but there is no prior work to integrate both of them for biomolecules detection. This invention combines photonic crystal with magnetic beads into a fluorescent immunoassay with extremely low limit of detection. With the light resonance of photonic crystal and high surface to volume ratio of magnetic beads, as well as the magnetic beads aggregation, this invention offers two advantages(1)fluorescence enhancement over thousands times (2)detection limit dramatically redued to sub-fg/ml level, which can not be achieved by the current beads-based immunoassay.
