技術說明

以palmitic和 lauric acid進行疏水性修飾低分子量乙二醇幾丁聚醣以形成具有親水性和親脂性的兩性聚合物LMWGCP和LMWGCL(low molecular weight palmitoyl glycol chitosan和low molecular weight lauroyl glycol chitosan),來提供做為輸送系統的藥物載體。 在以各種不同脂溶性及分子量的藥物(包括5-aminolevulinic acid (ALA, log K = -0.14), hydrocortisone (log K = 1.28), estradiol (log K = 3.84), curcumin (log K = 2.82), and PEG 4000)穿皮試驗下,LMWGCP促進了所有試驗藥物的皮膚滲透性,而LMWGCL則促進了前四種藥物的皮膚滲透性,但造成curcumin的緩釋效果;對於hydrocortisone和ALA的效果則以LMWGCP大於LMWGCL。兩種材料亦促進上述藥物在皮膚的吸收,具有發展為經皮和局部皮膚藥物傳輸載體的潛力。 Low molecular weight glycol chitosan was hydrophobically modified with palmitic/lauric acid to form polymeric amphiphiles consisting of both hydrophilic and hydrophobic properties. The potential of LMWGCP/LMWGCL as carrier for skin delivery of drugs with different physicochemical properties, including 5-aminolevulinic acid (ALA), hydrocortisone (HC), estradiol (ES), curcumin (CUR), and PEG 4000 through mouse skin in vitro. LMWGCP hydrogel enhanced skin permeability of all the drugs. LMWGCL hydrogel enhanced skin permeability of ALA, HC, ES and PEG 400 but sustained the skin delivery of CUR. The degree of enhancement for HC and ALA was greater in LMWGCP than LMWGCL. LMWGCL deposited more amounts of ALA, HC and PEG 4000 in the epidermis than LMWGCP. However, the deposition of ALA, HC, CUR and PEG 4000 in the dermis was greater from LMWGCP than LMWGCL. The LMWGCP/LMWGCL can potentially be applied as delivery vehicle to improve transdermal and topical delivery of various drugs.
