技術說明

本發明係關於一植物老化誘導啟動子，以及其相關之重組質體與轉植植物。基因的表現是調節細胞生理的重要步驟，目前用於植物基因表現的啟動子，大都是用菸草鑲嵌病毒 35S啟動子，35S啟動子可廣泛表現於植物各部分組織，對於需要進行組織特異性表現的基因，或是廣泛表現基因會對於植物有害時，35S啟動子便不適用，這時具有組織特異性表現的啟動子，將是最好的選擇。植物細胞老化時會啟動一連串基因以進行老化過程，因此本發明利用相減式雜合反應，分離出文心蘭花瓣老化時會大量表現基因，再由文心蘭的基因組中分離出此基因的啟動子，因此啟動子具有在植物老化時會大量啟動下游基因表現的能力，利用此啟動子之特性，可以在植物老化時啟動特殊基因，以用於防止植物老化，或延長植物老化過程，增加農作物附加價值。 The present invention relates to a plant senescence-inducible promoter and its relevant recombinant plasmid and transgenic plants. Gene expression is an important process to regulate cellular metabolism. Currently, most plant promoters, such as TMV 35S promoter, used for gene expression are constitutive active in all developmental stages. However, development-specific promoter is required for the expression of certain genes, which are harmful to plants, and can be used to induce the accumulation of gene product in the specific developmental status. The onset of senescence in plant cells includes a series induction of several genes. In this study, a senescence-inducible gene was isolated from the flowers of oncidium using subtraction hybridization, and its promoter was further obtained from genomic DNA. This specific promoter driving gene expression in the senescence stage of plant can be used to postpone the beginning of plant senescence, and is valuable to agriculture.
