細胞死活特性做為評估細胞生理狀態的重要標記,不單能用於衡量細胞現有狀態,更被用於評估細胞對外在環境刺激之反應,比如藥物耐受性、毒化物反應、氧化壓力與放射線暴露下之反應等等。此外,當細胞處於不同生理狀態時,其細胞質與細胞膜之電學特性(介電質與導電度)會出現明顯之差異,進而影響了細胞之極化能力。更進一步,細胞極化能力之差異會促成介電泳操作強度上的不同。因此,細胞間之介電泳強度可用以區別細胞之死活程度。再者,由於光誘導介電泳可使用投影機生成之光圖形來控制細胞之移動,因此在操作上較傳統金屬電極有更高之可控性與靈活度。同時,相較於光誘導介電泳常規使用之靜止或定速光圖形,本發明使用之變速度動態光圖形模組可以更好地區別不同之細胞死活程度,因而能提高細胞分離與純化之解析度,能大幅改進後續研究之精準度。總和上述,本發明作為一簡易、無標記且高解析度之細胞死活特性分離純化平台,可篩選出不同環境耐受性之細胞群體(如抗藥性、放射線抗性等),以供後續研究或臨床治療診斷所用。 Cellular live and dead properties are considered as a meaningful indicator to assess the physiological status of cells. It can not only be used to measure cells health and growth but also applied to evaluate the cell response to external environmental stimuli, such as drug resistance, toxicity reaction, oxidative stress and radiation resistance. In addition, when cells are in different physiological status, the electrical properties (permittivity and conductivity) of cell cytoplasm and membrane show significant variance, thus changing the degree of cell polarization. Furthermore, the discrepancy of cell polarization leads to different manipulating strength of dielectrophoresis. Therefore, the dielectrophoresis intensity of cells can provide a method to distinguish cells population expressing distinct cellular live and dead properties. Moreover, because optically-induced dielectrophoresis (ODEP) device can control the cell movement by using the light pattern generated by the projector, it is more controllable and flexible in manipulation than the conventional metal electrodes dielectrophoresis (DEP) device. Meanwhile, compared to the static or fixed-speed light patterns used in conventional ODEP manipulation, the accelerated-velocity dynamic light pattern model utilized in this invention can achieve the better live and dead cells distinguishment and increase the resolution of live and dead cell separation and purification, dramatically improving the precision and accuracy for follow-up research. Taken together, this invention display a simple, label-free and high-resolution ODEP platform for cell separation and purification based on cellular live and dead properties, utilized to screen and isolate the cell populations expressing various environmental tolerances (eg, drug resistance, radiation resistance, etc.), serves as a tool for follow-up study or clinical diagnosis and therapy.
