本發明為一種提升於外部電場下製作之蛋白質晶片效能之方法及由各方位(0°-360°)提供外部電場之裝置,其方法係於以不同角度施加之外部電場下製作各蛋白質晶片,再量測各蛋白質晶片上蛋白質分子與其配體分子間之結合力,即可推知此將蛋白質分子用於蛋白質晶片製作時以何種角度施加外部電場可使蛋白質晶片之效能提升至較佳或最佳,另,由各方位提供外部電場以製作各蛋白質晶片之裝置具有可環繞承載蛋白質晶片之載台轉動(0°-360°)之電場支架,電場支架上相對設置之電極可對載台上之蛋白質晶片提供不同角度之外部電場。 This patent elucidates the direction of immobilized proteins on chip which can be re-oriented by an external electric field (EEF) leading to the change of protein-protein interaction. . For this purpose, we design a set-up with (+)/(-) electrodes (producing EEF) included in a 360o rotatable disc. Protein chamber is located in the center of disc (equal distance between (+) and (-) electrodes). The binding forces (BF) between two proteins (anti-cancer peptide, CB1a, immobilized on AFM tip (afm_CB1a) and its anti-body, anti-CB1a, immobilized on chip (chip_anti-CB1a)) were investigated by using atomic force microscopy (AFM) at different EEF angles. Optimum binding interaction can therefore be obtained from the relationship of binding forces vs. EEF angles. Result showed that the effective interaction was increased to about 116.1%. This technique of optimization can be applied to the measurement of maximum protein-protein interactions, high efficient protein chip
