Polyacetylenic compounds for protecting against pancreatic islet atrophy

第二型糖尿病是一種涉及異常胰島素產生途徑與效能的嚴重慢性疾病。促胰島素分泌劑(secretagogue)藉由增進胰島素產生來治療第二型糖尿病。植物為提供第二型糖尿病藥物豐沛的天然物來源;我們從大花咸豐草中萃取出一種聚多炔糖苷,命名為cytopiloyne,在db/db小鼠或BALB/c小鼠具有降血糖的作用,此外我們發現cytopiloyne可以經由細胞的鉀離子通道及鈣離子通道,來刺激其分泌胰島素。根據這些觀察,進一步的實驗證明cytopiloyne會促進細胞的胰島素基因轉錄及胰島素的產生。我們的結果建議可以用cytopiloyne作為治療第二型糖尿病的藥物前驅物。此外這個化合物可作為咸豐草當作治療糖尿病的指標成分,作為品管使用。 We have found a fraction from B. pilosa that increases insulin production from βcells and decreases blood glucose levels on diabetic db/db mice. Furthermore, we have isolated a pure compound at 25 ug/kg BW with the same activity on diabetic db/db mice. Those data indicate that the fraction can be use as a botanical therapeuticals or alternative/supplementary medications of type 2 diabetes. And the pure compound from B. pilosa can be served as medications or lead drugs to cure type 2 diabetes. The effective dose of pure compounds for sugar lowering is thousands times lower than that of 2-D-glucopyranosyloxy-1-hydroxy-5(E)-tridecene-7,9,11-triyne and 3-D-glucopyranosyloxy-1-hydroxy-6(E)-tetradecene- 8,10,12-triyne.
