技術說明

本發明係揭露一種運用光纖式粒子電漿共振感測器之動力學常數估算方法, 其步驟主要包括: 提供光纖式粒子電漿共振感測器; 分別取得曲線關係圖中第一段落以及第二段落之反應初始之時間所對應之光訊號強度值、反應達到動態平衡時之光訊號強度值以及參考光訊號強度; 將取得之光訊號強度值帶入由準一級反應速率方程式假設下建構之模型所推導出之公式中。據此, 本發明係迅速注入待測溶液後於靜置狀態下量測光訊號強度隨時間之變化。不同於現有連續流動型式之電漿共振感測器, 在量測結合速率常數與分解速率常數時, 其上限不會受制於注入流率。 A method for obtaining the binding kinetic rate constants using fiber optic particle plasmon resonance (FOPPR) sensor, which employs the following major steps: providing one FOPPR sensor instrument system, obtaining optical signal intensities at the initial time and the steady state signal intensities of the first and second regions in the intensity versus time curve separately, substituting the measured signal intensity values into the formula which is derived using pseudo-first order rate equation model . According to this method, no fluorophore labeling is required. In addition, this method measures the temporal signal intensity evolution under static conditions as the samples are quickly loaded. As a result , unlike the conventional device where the sample is continuously infused, the method is able to measure the association and dissociation rate constants of which the upper bound are not limited by the sample flow rate.
