技術說明

一種自柔軟指形軟珊瑚分離出新化學結構之天然物 Thioflexibilolide A之方法包含步驟如下，利用丙酮字柔軟指型軟珊瑚進行萃取;利用二氯甲烷/水將萃取液進行分層萃取，取得二氯甲烷層及水層;將二氯甲烷層以管柱層析法進行初步分離並利用正己烷/乙酸乙酯/丙酮/甲醇進行初步分離，以取得數個劃分層提取液;在正己烷/乙酸乙酯比例為1:1的分層提取液利用反相高效能液相層析儀進行純化下取得Thioflexibilolide A，運用核磁共振儀確認Thioflexibilolide A為一種新結構之化合物。其中Thioflexibilolide A,在酯多醣刺激小鼠巨噬細胞(RAW264.7 cell line)模式下，具有抑制前發炎性蛋白質誘發型一氧化氮合成酶與第二型環氧化酶蛋白質之表現，同時在六羥多巴胺誘發人類神經母瘤細胞凋亡模式下，在Alamar blue assay與TUNEL assay的分析中，具有保護神經細胞之作用。 A new chemical structure of natural product, Thioflexibilolide A, isolating from Sinularia flexbilis, includes the following steps.Utilize acetone to extract solution. And utilize methylene chloride/water to partition two layers.Then, separate the organic layer by chromatography, and gradient elute the layer by n-Hexanes/EtOAc/acetone/MeOH. A fraction eluted with n-Hexane/EtOAc (1:1) is further purified by HPLC (MeOH/H2O, 2.3:1) to afford the compound . And using nuclear magnetic resonance to identify thechemical structure. And it significantly inhibited the expression of pro-inflammatory proteins(iNOS and COX-2)in lipopolysaccharide-stimulated murine (RAW264.7) macrophage cell. By using Alamar Blue assay and TUNEL staining, it also performed neuroprotection activity in 6-OHDA induced apoptosis in the human neuroblastoma (SH-SY5Y) cell line .
