技術說明

本發明為利用第二型膠原蛋白，處理於體外大量培養增生之軟骨細胞，使其再度表現軟骨性細胞標誌之方法。本發明之特點為可使體外培養擴增至已瀕臨靜止，放大至少第八代(放大38倍)之軟骨細胞，於分盤為第九代時再度表現內生性之第二型膠原蛋白，Sox9轉錄因子，與aggrecan核心蛋白，並增加醣胺聚醣(Glycoaminoglycan)的分泌等。如此即可藉由體外培養擴增方法，獲得數量足夠並具有生理活性之軟骨細胞，以應用於自體移植之軟骨修復治療。 After extensively expanded in monolayer culture, the chondrocytes become near quiescent and lose their chondrogenic characteristics. This invention is to establish a method that the phenotypic function of extensively expanded, near quiescent P9 primary chondrocytes after al least 11 times of cell doubling can be restored with type II collagen with or without IGF-I and TGF-1. As most of the related methods can only revive P4 primary chondrocytes after 3.8 times of doubling, this invention offers an efficient way to expand a scarcity of healthy chondrocytes for cartilage repair or regeneration.
