技術說明

提供一種基因剔除小鼠，其染色體上GNMT的基因座已被利用基因重組的方式破壞，其中該破壞位於序列識別號：8之547-4875，且該破壞造成一相較於野生型具有不正常肝臟功能之表現型。 本發明進一步提供一種篩選預防或治療肝癌或其他疾病之候選試劑(agent)的方法，其包含： 1.本發明之基因剔除小鼠 2.提供給該基因剔除小鼠一種候選試劑 3.比較已提供受試藥物及未提供候選試劑的基因剔除小鼠之肝臟功能，其中該候選試劑若能改善肝臟功能，則可被篩選出為預防或治療肝癌或其他疾病之有效試劑。 本發明進一步提供一組引子(primer)，其為序列識別號：1、2或是序列識別號：3、4。 本發明進一步提供一GNMT基因剔除小鼠之調節基因資料庫。 本發明進一步提供一肝癌之訊息傳遞基因資料庫。 The present invention also provides a method for screening a candidate agent for preventing or treating liver disease or disorder comprising: (a) providing the knock-out mouse of the present invention; (b) administering to said the knock-out mouse a candidate agent, and (c) comparing liver function of the knock-out mouse to that of the knock-out mouse of not administered said candidate agent; wherein the agent that ameliorates liver function is selected as an agent that has effectiveness against said liver disease or disorder. The present invention further provides a pair of prime, which is (i) SEQ ID Nos 1 and 2 or (ii) SEQ ID Nos 1 and 2. The present invention further provides a database for regulatory genes in GNMT knock-out mouse a database for regulatory genes in GNMT knock-out mouse. The present invention also provides a database for hepatocellular carcinoma signaling pathway genes.
