This method comprises the following steps: providing a bio-molecule combined with at least one ssDNA, wherein the ssDNA is combined with a first protein through an affinity binding tag; adding a second protein, the concentration of which is greater than that of the first protein; and replacing the first protein on the ssDNA with the second protein through the chemical competitive principle.
