胰臟癌之血清生物檢測標誌及其應用Methods for detecting pancreatic cancer

本發明係關於一種診斷胰臟癌之定量分析方法。我們發現一種蛋白質- UL16結合蛋白2 (UL16 Binding Protein 2, ULBP2, 又稱為NKG2D) - 過量表現於胰臟癌組織之腫瘤細胞中。我們同時進一步利用bead-based 免疫分析法進行定量實驗,發現此蛋白質含量在胰臟癌病人血清中較健康對照組血清中顯著升高,可以用來做為診斷胰臟癌之血清生物標誌。且同時測量胰臟癌病人血清中的CA19-9表現量,結果顯示ULBP2在胰臟癌病人血清中過量表現可以提高使用CA19-9做為胰臟癌病人臨床診斷的偵測效力。我們在大腸直腸癌、鼻咽癌與胃癌的病人血液中偵測ULBP2的含量,發現ULBP2含量在胰臟癌病人血液中較這三種癌症病人血液中顯著升高。 關鍵字:ULBP2、胰臟癌、bead-based免疫分析法、血清生物標誌 We identified UL16 binding protein 2 (ULBP2) as a protein overexpressed in pancreatic cancer tissues. We developed a bead-based immunoassay to measure the ULBP2 level in serum samples from pancreatic cancer patients and healthy controls, and found that ULBP2 levels were significantly higher in pancreatic cancer patients than those in healthy controls. This invention provides a method to detect pancreatic cancer using ULBP2 as a serological marker. We also measured the serum CA19-9 levels in the same cohorts and found that combination of ULBP2 and CA19-9 promotes the efficacy of pancreatic cancer detection. We measured the blood ULBP2 levels in patients with other cancer types, including colorectal carcinoma, nasopharyngeal carcinoma and gastric cancer. We found that blood ULBP2 levels were higher in patients with pancreatic cancer than other cancer types. Keywords: ULBP2、pancreatic cancer、bead-based immunoassay、serum marker
