技術說明

本發明提供檢知一待測物的比色免疫檢測方法，包含下列步驟:在一基材表面聚合一多孔性薄膜層。使標定物吸附於多孔性薄膜層。將一金奈米粒子與一二級抗體結合。加入具有待測物的待測液體，使金奈米粒子、二級抗體及待測物結合成一檢測組偶聯物。將檢測組偶聯物導入多孔性薄膜層，使待測物與標定物專一性地結合，若待測液體含有待測物時多孔性薄膜層呈現一預定顏色，若待測液體不含有待測物時多孔性薄膜層則無顏色變化。最後觀察多孔性薄膜層顏色變化的長度來判讀抗原濃度。本發明另亦提供一種檢測待測物之比色免疫裝置。 The invention relates to a method of colorimetric immunodetection. The monoliths are first photopolymerized within a substrate surface. Then the monoliths are modified with primary antibodies. Gold Nanoparticle (AuNP) colloids are conjugated with secondary antibodies. Adding an antigen and the secondary antibody specifically binds the antigen. The AuNP, secondary antibody and antigen incubate into detection conjugate. The detection conjugate is then injected into the monoliths, and the primary antibodies specifically bind to the antigens. Finally the amount of antigens can be determined via the color variance of the bound AuNPs label on the monolith.
