技術說明

本發明揭示一種化合物的用途，其為用於製備哺乳類TOR抑制劑，其中化合物為corylin。 我們利用酵母菌建立一套抗老化藥物的快篩系統，並利用此快篩實驗平台發現補骨脂的酒萃物能有效增加酵母菌的壽命。接著也在三向顯微鏡定位系統中，觀察到補骨脂確實能延長酵母菌的replicative life span (RLS)，且是經由抑制mTOR路徑達到延緩老化的目的。不但如此，將補骨脂的酒萃物利用管柱層析法提取各種純化物並分析後，我們發現Corylin能延長酵母菌的RLS，而且也是經由抑制mTORt的路徑達到延緩老化的目的。 The invention of a compound, and the use is for the manufacture of mammalian TOR inhibitor, the compound being:corylin. The budding yeast Saccharomyces cerevisiae has been used extensively to study the fundamental processes of replicative life span (RLS). Due to the inherent difficulty of the RLS assay, it is inefficiency to screen drugs for extending RLS in yeast. Here, we developmented a high-throughput drug screening for increasing RLS in yeast. By using the system, we found that the ethanol extract of Fructus Psoraleae extended replicative life span in yeast. Following a column chromatography procedure, we analyzed the compounds that isolated from the ethanol extract of Fructus Psoraleae for increasing life span. We determined that Corylin extended replicative life span by the inhibition of mTOR pathway.
