利用屏蔽效應檢測多巴胺及其檢測試劑Utilization of Shield Effect for Dopamine Detection and Reagent Development

本發明係利用可與多巴胺結合之人類單胺氧化酶於特定波長所特有的吸收光譜特性,經由單胺氧化酶蛋白-綠色螢光蛋白所融合之探針蛋白產生屏蔽效應的影響,當未結合多巴胺等人類單胺氧化酶受質時,能阻擋綠色螢光蛋白的螢光訊號,而於多巴胺等受質進入作用時,綠色螢光蛋白的螢光訊號能被釋放,進而使螢光探測儀器測定。藉由此釋出之綠色螢光偵測細胞和組織在生理狀態下及檢體中的多巴胺濃度和變化,並發展成可偵測多巴胺的探針,更進一步成為特定的檢測探針試劑。 This invention attempts to engineer a “dopamine probe” that could detect dopamine. The human monoamine oxidase B (MAO B) is selected as probe for its binding specificity to dopamine. Previous studies showed that oxidized form of MAO B-FAD presented special spectral absorption. Such spectral property dismissed after MAO B-FAD reduction by binding to substrates. Taking this unique disposition of MAO B, this invention generates fusion proteins of MAO B-green fluorescent protein (GFP) to be used for dopamine detection through the “shield effect”. This innovation is aim to apply in both basic research and clinical study.
