超順磁氧化鐵奈米團簇之製備方法

本發明係提供一種製備水溶性超順磁氧化鐵奈米團簇之方法,其步驟包含:a)將超順磁氧化鐵奈米粒子及生物相容性高分子溶於有機溶劑中形成有機相溶液;b)將該有機相溶液加入水相溶液中,並使用超音波分散進行乳化以形成超順磁氧化鐵奈米團簇;以及c)去除該有機溶劑。藉著超音波乳化法及有機溶劑揮發法,以製備水溶性超順磁氧化鐵奈米團簇,可有效降低製備時間並簡化製備程序,且增加於水溶液中之單分散性及穩定性的功效。此奈米團簇的顯影效果較市售的顯影劑效果好。 Superparamagnetic iron oxide nanoparticles (SPIO) has been used as T2 magnetic resonance contrast agents clinically by reducing spin–spin relaxation times in water. To prepare magnetic clusters, several kinds of surfactants were used to convert the hydrophobic SPIO from oil phase to water phase and alos stabilize the cluster structure in aqueous solution. We use a biocompatible polymer for stabilization of oil phase SPIO in the emulsion process. The alkyl chains of PEG were wound around the SPIO to stabilize the oil drop and the hydroxide group of PEG helped the oil-drop well dispersion in the aqueous solution after sonication. The resulting SPIO clusters are uniform with a size less than 200 nm that revealed excellent long-term stability in the aqueous solution for one month. The proposed procedure of SPIO cluster formation is efficient and time-saving with high yield comparing to the previous reports.
