技術說明

本發明之主要目的在於提供一種具有脂多醣結合活性之胜肽及其於製備抗生素、飼料添加物之用途，以及內毒素移除、檢測之套組與方法。本發明所述之具有脂多醣結合活性之胜肽，其中該胜肽係源自於淡水長腳大蝦(Macrbrachium rosenbergii)的抗脂多醣因子。 本發明所述之具有脂多醣結合活性之胜肽，其中胜肽具有抗微生物之活性，且該微生物可為革蘭氏陽性菌、革蘭氏陰性菌、酵母菌(Pichia pastoris)、乳酸球菌(Lactococcus garvieae)、金黃色葡萄球菌(Staphylococcus aureus)、大腸桿菌(Escherichia coli)、霍亂弧菌(Vibrio cholerae)或產氣單胞菌(Aeromonas hydrophila)。 內毒素移除方法，包含有下列步驟：(a)固定一如前所述之胜肽於一固相擔體；(b)提供一樣本溶液與該胜肽反應；以及(c)收集與該胜肽反應後之樣本溶液。內毒素檢測套組，包含有下列步驟：(a)固定一如前所述之胜肽於一固定基質；(b)提供一脂多醣分子與該胜肽反應；(c)提供一偵測分子與被捕捉的該脂多醣分子反應，其中該偵測分子具有一標定物以及一如前所述之胜肽；以及(d)測定該標定物。 The finding primarily supplies a type of LPS-binding peptides to being used as antibiotic and a feed supplement for aquiculture, and to produce a kit of LPS assay and LPS removal. These peptides were derived from anti-LPS factor of Macrbrachium rosenbergii。 One of the LPS-binding peptides has antimicrobial activity to inhibit the growth of Gram positive bacteria Lactococcus garvieae and Staphylococcus aureus, Gram negative bacteria Escherichia coli, Aeromonas hydrophila andVibrio cholera. For the removal of LPS, main steps of the procedure include (a) peptide fixation, (b) one LPS-containing sample loaded and binding the peptide, and (c) collection of peptide-bound sample. As for the kit of endotoxin assay, main steps of the procedure are (a) LPS-binding peptide fixed on a solid phase, (b) LPS-containing sample loaded to react with peptide, (c) supplement of the fluorescein-labeled LPS-binding peptide, and (d) determination of fluorescent strength.
