技術說明

脂肪幹細胞(Adipose derived stem cells, ADSCs)已被認定是一種可應用於成體幹細胞或出生後之先驅細胞的來源。ADSCs被認為有一些比骨髓間質幹細胞(Bone marrow mesenchymal stem cells, BMSCs)更有利於應用之處，包括：容易分離、數量較多以及在培養中容易增生。然而，在活體外長期培養放大ADSCs之量有被病毒感染或細胞變性之可能性，這些將會阻礙ADSCs之應用於細胞治療。因此，我們認為最安全的方式是在手術當中應用一個無菌過程有效的分離脂肪中之ADSCs，立即自體移植作為細胞治療。本案旨在開發一個短時間且簡易流程進行脂肪幹細胞分離與純化開發全新醫療儀器經無菌過程有效的由皮下脂肪分離及篩選夠量且的ADSCs進行再生醫學的組織重建。預期在幹細胞運用於再生醫學上達到快速安全的效果。本發明揭露能夠從脂肪組織中取得幹細胞之自動化系統，其能夠於同一裝置中以自動化方式進行脂肪組織分散、細胞收集，以及幹細胞篩選。 Adipose derived stem cells (ADSCs) have been identified as an alternative source of adult stem cells or postnatal progenitor cells. ADSCs are also thought to have several advantages over bone marrow mesenchymal stem cells (BMSCs), including ease of isolation, relative abundance, and rapidity of expansion in culture. However, long term in vitro culture of ADSCs increases risks of contamination and transformation. Therefore, we propose that the safest way to use hADSCs for cell therapy is to harvest ADSCs for autologous transplantation during surgery with quick and efficient process. We develop a short-time and simplified protocol for ADSCs isolation from subcutaneous fat and selection with an aseptic process medicine. All the isolation processes are included by automatic system, that is, isolation, collection and selection of ADSCs.
