技術說明

本技術所使用微衛星分子標誌(microsatellite markers)，又稱簡單重複序列(simple sequence repeat, SSR)，具有高度多型性、高再現性、易以PCR反應偵測、操作相對快速簡易、僅需少數植物樣本、不受植物生長環境與時期影響且可適用於高通量分析等優點，可為一穩定、迅速且可靠的蝴蝶蘭品種鑑定技術。目前已發展10組螢光標定之微衛星分子標誌，以自動化核酸定序進行基因型分析，鑑別 12個常用於育種上的蝴蝶蘭原生種，結果顯示只需少數幾組微衛星分子標誌即可鑑別不同蝴蝶蘭原生種。根據每個原生種獨特的DNA分子條碼，建立蝴蝶蘭原生種DNA指紋分析系統。未來可應用於評估蝴蝶蘭品種所含有之物種組成，應用於鑑定蝴蝶蘭商業栽培種的親緣分析。 Microsatellites, also called simple sequence repeat (SSR), are widely used as powerful tool for Phalaenopsis variety identification because they are highly polymorphic, highly reproducible, easily detected by PCR reaction only small amount of plant material needed and being amenable for high-throughput analysis. A total of 10 fluorescence-labelled microsatellite markers with automatic DNA analyzer have applied to discriminate 12 Phalaenopsis wild species. The results showed that few microsatellite markers were enough to discriminate the Phalaenopsis wild species mentioned above. Species-specific DNA molecular identity was generated and collected to establish the DNA fingerprinting system.
