羥基苯甲酸酯 (para-hydroxybenzoate，又稱parabens) 是對羥基苯甲酸 (para-hydroxybenzoic acid，簡稱PHBA) 的烷基酯類，為一類廣泛使用的防腐劑，經常添加在食品、化妝品及藥品中，常見的包括甲基(methylparaben，簡稱MP)、乙基(ethylparaben，簡稱EP)、丙基(propylparaben，簡稱PP)、丁基(butylparaben，簡稱BP)。早期研究Parabens可快速吸收及代謝成無毒性的PHBA，且無致突變性、致畸性、致癌性。但近年來逐漸有研究提出parabens具有雌激素(oestrogen)的活性及會影響男性生殖系統，被歸類為仿雌激素(xenoestrogens，簡稱XEs) 和內分泌干擾物(endocrine-disrupting compounds，簡稱EDCs，又稱作環境賀爾蒙)，可能對人體產生健康方面的危害。Parabens可透過食入、吸入或皮膚接觸等方式進入體內，其中透過化妝品及藥品的途徑接觸到parabens的比例較高，且多項文獻指出parabens(尤其MP)會引起過敏性反應及光敏感性。有鑑於parabens的廣泛使用，本技術旨為設計一套簡單、快速的分析方法透過介質輔助雷射脫附游離飛行時間質譜儀 (matrix-assisted laser desorption ionization time-of-flight mass spectrometry; MALDI-TOF MS) 配合化學衍生方法來同時檢測並加以定量市售產品中常見四項parabens的含量。結果指出本方法對於四項parabens (MP、EP、PP、BP)皆有良好的線性關係，定量範圍為0.1-10 ug/mL，決定系數(coefficient of determination，R2) ≥0.9995，相對標準偏差(relative standard deviation，RSD) <9.86%。本技術可應用在檢測液狀(solution)、乳狀(lotion or emulsion)、霜狀(cream)等不同質地的樣品，是一套快速、簡單的對羥基苯甲酸酯類防腐劑之分析及定量方法。 Parabens are esters of para-hydroxybenzoic acid, which are widely used as preservatives in food, cosmetic and pharmaceutical products. They are effective antimicrobial agents, with activity increased with the increasing length of the alkyl chain, include methylparaben (MP), ethylparaben (EP), propylparaben (PP) and butylparaben (BP). Parabens are rapidly absorbed and metabolized into their common metabolite (para-hydroxybenzoic acid), a non-toxic product. Recent years, parabens have been shown to induce allergy reaction, possess estrogenic activity and affect male reproductive system. The results of the method showed the quantitative range of four parabens were 0.1-10ug/mL with the coefficient of determination (R2) ≥ 0.9995 and relative standard deviation (RSD) < 9.86%. A rapid, simply analytical method for simultaneous determination of four commonly used antimicrobial preservatives, para-hydroxybenzoate (parabens), in different formulation samples was established.
