技術說明

微針貼片是將傳統的針劑注射與經皮貼片之優點相結合的一種無痛、微侵入式醫療裝置,利用微米級之小針可直接刺穿皮膚角質層,不刺激位於真皮層的神經系統,有效地將大分子藥物傳送至皮膚內。目前所開發之高分子微針貼片,皆屬快速釋放藥物劑型,尚無可持續釋放藥物之長效劑型。本研究將幾丁聚醣溶液以離心方式灌模至半乾後,再與聚乳酸[poly(L-lactide-co-DL-lactide), PLA]支持軸進行組合,製備出一可鑲嵌式經皮微針貼片。此微針可經由徒手施壓聚乳酸支持軸方式,成功刺入皮膚並與支持軸分離,鑲嵌於皮膚中持續地釋放藥物(F001)。由豬皮穿刺實驗證實,此微針確實可成功完全刺入豬皮中,直達真皮層(~500 um)。此緩釋型微針貼片,可減少病患之施藥次數並增加用藥之便利性,避免貼片長時間黏貼於皮膚所造成之不適。目前正與成大微免所與生化所合作,共同開發成為緩釋型經皮疫苗貼片,將疫苗標的傳輸至富含抗原呈現細胞的表皮層與真皮層之間。幾丁聚醣微針除了具有緩釋疫苗的效果外,材料本身還有免疫佐劑的特性,將可提升疫苗之功效。 We developed an embeddable transdermal microneedle device, which combined chitosan microneedles with a poly(L-lactide-co-DL-lactide)(PLA) supporting base array. To prevent damaging the integrity of encapsulated biomolecules, a gentle fabrication process for chitosan microneedles was developed. First, viscous chitosan solution was centrifuged at ambient temperature to fill the microneedle mold. Subsequently, a PLA supporting base array was aligned to the filled cavities to make a complete microneedle device. In vitro porcine skin insertion test showed that the chitosan microneedles can separate from the PLA supporting base and be left in the skin and the insertion ratio reached almost 100%, respectively. The histological results also indicated that the chitosan microneedle can be inserted into the dermis layer and remain within the skin for sustained drug release.
