技術說明

抗體藥物或可溶性受器透過專一的中和疾病區域抗原達到療效，目前已廣泛應於治療多種疾病，2010 全球十大藥物即有五項為抗體藥物，2015 年全球市場可望超過 600 億美元。但許多抗體藥物的辨認抗原除在疾病區表現外，亦會於正常組織表現，使得抗體藥物長期使用易產生副作用。因此提升抗體對疾病區抗原的選擇性，而不去抑制正常組織的抗原實為當務之急。本發明創新利用各種抗體自身腰部之 hinge 結構做為抑制區將其移植的抗體頭部結構，藉由 hinge 屏蔽抗體結合位，並使用蛋白酶專一性受質胜肽連結抗體和抑制區，形成前驅抗體藥物，使前驅抗體唯有在蛋白酶過度表現的疾病區域，才能被水解並移除抑制區，回復抗體與抗原的結合能力，並選擇性中和疾病區抗原，提高選擇性標靶治療，降低副作用。目前此創新的閉鎖器已可成功應用於多種抗體上，如:anti-TNF-a antibody (REMICADE .HUMIRA)、anti-HER2 antibody、anti-EGFR antibody及 anti-RANKL antibody (PROLIA)，閉鎖器可有效的抑制這些抗體與抗原的結合能力，唯有在蛋白酶作用後，才能夠回復其功能。同時我們也證實了各種亞型抗體的 hinge 結構皆可做為閉鎖器。結果顯示此閉鎖器相當具有可行性，且具有以下優點:1.具廣泛應用性。2.不會引起任何免疫反應。3.提高抗體對疾病區域選擇性。相信本閉鎖器的成功，將有效的提高現有標靶抗體藥物對疾病區域的專一性，特別是慢性疾病的終身用藥，可降低對正常組織區域抗原的結合，進而提高抗體藥物的選擇性，並減低其副作用。 Ab drug is widely used to cure many diseases, however, the Ab treatment presented can’t distinguish the target antigen in disease region from normal region which causes systemic targeting and induces the side effects. Consequently the enhancement of Ab-specificity to disease region is critical important. Our strategy is use of protease substrate peptide as the linker to combine the inhibit domain (hinge) and Ab binding site to generate the protease-activated pro-Ab. The hinge domain will cover the Ab-binding site inhibiting binding capacity of Ab. The protease substrate peptide can only be hydrolyzed by protease which over-expressed in the disease region, then the Ab can specifically target to the disease region to improve the selectivity of disease target therapy. In previous results, we have already confirmed that the hinge domain could reduce the binding capacity of REMICADE, HUMIRA, anti-HER2 Ab, anti-EGFR Ab, and PROLIA.
