技術說明

尿液的取得，對於甲狀腺癌的病患而言，是不具侵襲性，尿液中之細胞外泌體(Exosome)是癌症預後指標及治療的重要因子和標的。我們進行尿液中之細胞外泌體(Exosome)生物標記分析。經由AB SCIEX QTRAP 5500 LC/MS/MS System 配合 Multiple Reaction Monitoring (MRM) 技術針對特定蛋白質序列片段進行定量分析。發現在接受甲狀腺切除手術後尿液的細胞外泌體(Exosome)內甲狀腺球蛋白(Thyroglobulin)與半乳糖凝集素-3(Galectin-3)有高濃度的偕同上升現象，相較於目前臨床報告，經甲狀腺切除手術及經放射碘-131治療患者的血液中甲狀腺球蛋白(Thyroglobulin)更易於術後追蹤時於早期被偵測出。 We use peptide sequence to quantify the levels of thyroglobulin in urine exosome. Significant reduction in cost-spending in rhTSH consumption, and patients do not need to stop using thyroid hormone during the course of cancer follow-up. We also use galectin-3 as a comparator to confirm the trends of thyroglobulin levels. Galectin-3 could act to be the modulator of cell growth via galactoside-binding protein, correlating to occurrence and metastasis of papillary thyroid carcinoma.
