技術說明

軟骨合成與分解的不平衡是造成關節軟骨退化病程發展的主要因素, 然而以放射顯影的偵測，其限制是軟骨退化已經有顯著的破壞了才可以偵測到，病患在發病早期無法檢測出來。一些衍生於骨頭、軟骨與關節滑液中的生物標誌也常被選擇來偵測關節軟骨退化的進展，但大部只能檢測血清與關節滑液之檢體。為了避免用侵入檢測方式並且能快速檢驗，因此本專利的主要目的為以尿液檢體為主，研發快速簡易的檢測系統。一種用以生產第Ⅱ型膠原片段抗體的融合瘤細胞株，該融合瘤細胞株係寄存於新竹食品科學工業發展研究所，寄存編號為BCRC 960423，該抗體係專一性結合於一第Ⅱ型膠原片段，該第Ⅱ型膠原片段係如SEQ ID NO：1所示之胜肽片段；一種以尿液檢測退化性關節炎之方法，係利用該抗體與尿液中退化性關節炎之生物標記結合，該生物標記係包含第Ⅱ型膠原片段 11個胺基酸。 Articular cartilage degeneration is a complex heterogenous disease. The imbalance in cartilage synthesis and degradation is major to the development of articular cartilage degeneration. To prevent from invasion to patients bodies to collect the specimens, the purpose for this project will develop the rapid detection system to exam the urinary samples from articular cartilage degenerative patients. A hybridoma cell line for producing an antibody of Type Ⅱ Collagen Neoepitope (TIINE), which is deposited at Bioresource Collection and Research Center of Taiwan with a deposit number BCRC 960423. The antibody has the capability to couple with TIINE, and the TIINE is described as SEQ ID NO: 1. A detecting method for degenerative arthritis with urine sample, is by using the antibody mentioned above to couple with a biomarker of degenerative arthritis. The biomarker comprises a fragment of TⅡNE, which containing 11 amino acids.
