技術說明

一種無血清培養於高分子量透明質酸奈米塗層之細胞球體形成方法，係以無血清方式於透明質酸之奈米地形培養角膜基質細胞，高分子量透明質酸攜帶較多負電荷，當製備成奈米塗層同時具有較高表面粗糙度和高親水性之性質，實驗結果顯示細胞培養於透明質酸塗層(1.1-1.7nm)有助於增加細胞與細胞之作用，故有利於細胞聚集成細胞球體且維持於靜止生長之狀態，當細胞形成三維之細胞球體具有較高之生物合成能力並且能分泌更多細胞外間質，相當具有角膜基質組織修復應用潛力。 The invention provides a method of fabricating multicellular spheroids from hyaluronic acid coatings under serum-free cultivation. In one embodiment, a method of forming keratocyte cell spheroids is provided, which comprises culturing corneal keratocytes in a culture vessel coated with high molecular weight hyaluronic acid (≥ 360 kDa) under serum-free condition. The aim of this method was not only to maintain the cell viability and the proper phenotypic expression of the corneal keratocytes but also to exhibit greater amounts of collagen and GAG production. The fabricated bioengineered multicellular spheroids from this invention hold great potential for corneal stromal tissue reconstruction.
