技術說明

本發明係提供一種使用肺炎鏈球菌神經胺酸酶作為診斷和預防肺炎鏈球菌感染症之方法，主要是關於疫苗的開發，來預防肺炎鏈球菌的感染，以及診斷來自尿、血及膿胸的胸腔積液等檢體中是否感染肺炎鏈球菌。根據本發明研究，在肺炎鏈球菌引起溶血性尿毒綜合症（HUS）例子中，大部分肺炎鏈球菌都帶有三種神經胺酸酶基因(nanA、nanB和nanC基因)，雖抗NanC血清抑制或中和神經胺酸酶的活性並不如抗NanA和抗NanB血清，但將三種神經胺酸酶組合一起時則是最好的疫苗試劑，可作為一個理想的疫苗應試物，提供最好的保護效果來對抗肺炎鏈球菌的感染，達到良好之功效者。 A method of providing protection against pneumococcal infection in a subject is disclosed. The method includes steps of administering to the subject a composition that includes one or combination of three recombinant pneumococcal neuraminidases: NanA, NanB, and NanC of S. pneumoniae strains CGSP14, wherein administration of the recombinant pneumococcal neuraminidases elicits an immune response to S. pneumoniae, and treats the subject. In one embodiment, the method further includes a step of adding adjuvants to enhance the immune response. The method also includes a step of using passive antibodies, wherein said passive antibodies are anti-neuraminidase antibodies generated from neuraminidases: NanA, NanB, and NanC. Meanwhile, this invention also provides a method for the molecular diagnosis of pneumococcal infection.
