技術說明

We have constructed a multiplex oligonucleotide synthesizer capable of producing 24, 48, 96, or 384 oligonucleotides by microplate format at a time. The synthesis format and the number of channel are easily changeable. With the 384-channel format, 384 oligonucleotides of 20-mer in length can be synthesized in less than 10 hours. For each oligonucleotide, 5-20 nmol is prepared. Currently the synthesis step yield is greater than 99 %. The product purity and quality are evaluated by CE and UV-Vis spectrophotometer. The accuracy of the oligonucleotide synthesis has been verified by PCR and Southern hybridization experiments.
