技術說明

於一般攜載藥物的高分子微球核心表面以共沉法同時沉積具生醫相容性與血液相容性的無機殼層,並使具超順磁性(superparamagnetic)的奈米顆粒妝點(decorate)於殼層上。無機殼層為可透過水溶液化學法合成之生物相容性(包含血液相容性)陶瓷材料,主要功能在增加背景滲漏抵抗能力,並提供載體對高頻超音波的敏感性。超順磁性奈米顆粒為可透過常溫水溶液化學合成之生物相容超順磁性材料,可提供磁振影像T2 造影對比功能。本載體之無機殼層在無超音波作用時可阻止藥物外洩,於超音波轟擊後可因無機殼層的硬脆本質產生快速破裂,並阻止高分子的再組裝行為,因此可快速將藥物釋出; 無機殼層上所妝點的超順磁奈米顆粒在載體未被觸發前可發揮MRI T2造影對比功能,在超音波觸發之後,載體破裂造成超順磁性奈米顆粒分散程度改變,使磁場不均勻性產生變化,因此在超音波作用前後會有T2與T2*對比訊號的差異,可做為載體是否被觸發的判別參考。 In the present study, the superparamagnetic iron oxide (SPIO) nanodots were semi-embedded onto the surface of hydroxyapatite (HA)-coated liposomes to form a core-shell structure decorated with SPIO nanodots (i.e., HA/SPIO-coated liposome) which is expected to exhibit ultrasonically triggered behaviors and MR image contrast. The background leakage was significantly reduced by HA/SPIO coating layer. In addition, the HA/SPIO coating layer could enhance the US sensitivity of the novel drug vehicles in the high-frequency range (diagnostic US frequency range; 1–3 MHz). On the other hand, the ultrasonically triggered vehicle could exhibit a concentration-dependent T2 contrast in MR images. Most importantly, the r2* - r2 value of HA/SPIO-coated liposomes decreased after sonication. This suggests that the proposed vehicle could be used not only as a MR image-guided drug vehicle capable of ultrasonically triggered release but also as a MR reporter to probe ultrasonic triggering.
