技術說明

天然海洋化合物構成的豐富資源成為人類疾病的藥物的發展中扮演重要的角色。海洋衍生物WA-25為從澳大利亞軟珊瑚中萃取衍生而來，先前的研究已發現具有抑制血管新生的功能，並且是經由降低內皮細胞中的血管內皮增生因子及血管內皮增生因子受體2。本篇研究主要是在利用WA-25對抗腫瘤生長，並期望在未來可以有據有治療肝癌的潛力。研離體模式的結果顯現WA-25有效的抑制了肝癌細胞的生長侵入性及群落生長，並且造成癌細胞的凋亡。同時也抑制了癌症幹細胞自我更新及抗藥性的功能。治療的效果評估是經由原位肝癌動物模式和組織染色的學習。七天的治療顯示，WA-25可以有效的治療腫瘤的生長。結果發現WA-25有效的抑制肝癌的生長經由血管新生及癌症幹細胞，推測WA-25可以在未來成為有效的治療肝癌用藥。 WA-25 is a synthetic derivative during total synthesis of austrasulfone, a marine compound isolated from soft coral Cladiella australis. In this invention, we evaluated the anti-neoplastic efficacy of WA-25 for liver cancer. It was found that WA-25 inhibited the proliferation, invasiveness, anchorage-independent growth of human HCC cells in a dose-dependent manner. Besides, WA-25 induced apoptosis and attenuated the self-renewal capabilities in human and rat HCC cells. In Novikoff hepatoma model, WA-25 therapy significantly reduced the size and tumor burden of hepatoma in rats. Histological analysis revealed that WA-25 treatment increased the apoptotic TUNEL staining while decreasing the proliferating Ki-67 and neovascular CD31 expression. Moreover, WA-25 therapy significantly depleted the abundance of CD133+/CD44+ hepatic cancer stem cells in Novikoff hepatoma. Together, these results indicate WA-25 may facilitate a novel therapeutic alternative for HCC.
