苯醌類化合物應用於抑制黑色素瘤細胞之Wnt/β-catenin 訊息途徑之用途

近年保健食品開發,具臺灣本土性食藥用菇-樟芝甚受注目。我們過去研究發現,樟芝會誘導皮膚癌細胞(B16F1/B16F10)之凋亡作用。樟芝可利用現代生物栽培技術,生產液態深層培養之樟芝菌絲體發酵液,此方法最經濟且符合環保的人工培育法,可避免森林生態破壞、具有降低成本及大量生產的優點。本專利以皮膚癌細胞株(B16F10 and B16F1),作為樟芝菌絲體發酵液活性成分預防皮膚癌的標的細胞,發現具有多重抗皮膚黑色素癌包括抑制癌細胞增生、促進癌細胞凋亡、抑制癌細胞轉移及分子機制探討;動物實驗發現,樟芝可顯著抑制裸鼠皮膚癌腫瘤。 Antrodia camphorata is well known in Taiwan as a nutritional food/ beneficial mushroom. The aim of this study is to investigate AC-0, isolated from the fermented culture broth of Antrodia camphorata can inhibit melanoma progression (B16F10 and B16F1) via suppressed Wnt/-catenin signaling pathway. AC-0 showed a cytotoxic effect on both B16F10 and B16F1 cells determined by MTT and colony formation assay. We observed that AC-0 caused the G1 phase arrest. In addition, treatment of melanoma cells with AC-0 resulted in a significant increase in apoptosis. We also demonstrated that non- or sub-cytotoxic concentrations of AC-0 markedly inhibited the migration/invasion of highly metastatic B16F10 cells. These results suggested that AC-10 may exert anti-tumor activity by inhibiting Wnt/β-catenin pathway.
