利用生物反應器,培養植物細胞生產皮膚抗老化活性原料(EJE),並從中純化出單一活性化合物,進行抗皮膚老化之效用及分子生物活性評估。結果顯示:生物反應器中細胞培養液的磷、碳、鈣離子等濃度與含糖量及導電度方面與細胞生長代謝有高度的相關性;測量導電度則可作為評估細胞生長情形的指標。植物細胞生長曲線的建立有益於評估平行放大生產。抗老化活性的體外試驗結果顯示:EJE與單一活性化合物對膠原蛋白酶具有抑制性;在人類皮膚纖維母細胞實驗中,單一活性化合物經由MAPK pathway及AP-1抑制MMP-1生合成與促進Procollagen Iα1基因轉錄表現。單一活性化合物有延緩老化而促進細胞膠原母細胞生成之效用。動物實驗中,無毛鼠背部皮膚塗抹5% EJE的乳液(0.2ml/隻)可抑制因長期照射UVB所導致的皮膚增厚與皺紋生成等老化現象;塗藥組的組織切片也呈現具有改善無毛鼠之膠原纖維斷裂情形等。動物試驗印證5% EJE對於保護皮膚抵抗長期UVB曝曬所造成的氧化壓力及光老化等具有實質改善效用。 This study was focused on investigating plant cell produced antiageing component (EJE) applied in skin. In culture media.the ions varied in media, and further purified single compound from cultured cell to evaluate the anti-ageing efficancy in skin. In vitro, EJE and single compound exhibited the inhibition for collagenase. Besides, single compound also showed the decrease of MMP-1 expression through MAPK pathway and AP-1, but enhanced the gene transcription of Procollagen Iα1. All of that told the potentiality of single compound promised for prolonged ageing and procollagen produced. In vivo, the topical application of 5% EJE lotion (0.2 ml/mouse), the histology was showed the improvement of collagen fibers damage, the inhibition of MMP-1 expression and the reduction of hydrogen peroxide production by 5% EJE treatment in UV-induced hairless mouse skin. Both of results were supposed that 5% EJE was effective in elimination oxide stress and in improvement of photoaging.
