技術說明

本發明揭露一種可選殖啟動子的慢病毒載體，其包括：多選殖區，具 有選自由Cla I、Mlu I、Pst I、Sal I、Sma I 及Xma I 所組成之群組的複數個 限制酶切點，以選殖第一啟動子，其中第一啟動子可為持續型啟動子或誘 導型啟動子；報導基因，位於多選殖點之下游，當第一啟動子活化時，表 現報導基因所編碼之螢光蛋白質、冷光蛋白質或多胜肽；第二啟動子，其 為人類磷酸甘油酸激酶啟動子；哺乳動物篩選標記，其係位於第二啟動子 下游；以及細菌篩選標記。 A lentiviral vector for cloning promoter is disclosed. The vector comprises: a multiple cloning site having a plurality restriction enzyme cutting sites selected from the group consisting of Cla I, Mlu I, Pst I, Sal I, Sma I and Xma I for cloning a first promoter being a consecutive promoter or an induction promoter; a reporter gene located downstream of the multiple cloning site, a fluorescent protein, a chemiluminescent protein or a polypeptide encoded by the reporter gene is expressed when the first promoter is activated; a second promoter being a human phosphoglycerate kinase promoter; a mammalian cell selection marker located downstream of the second promoter; and a bacteria selection marker.
