運用一對離軸拋物面鏡以建構表面電漿波生醫檢測系統,此系統可做為對不同檢測方式的通用平台,運用拋物面鏡來建構的系統具有可調整的光入射角,同時保持光源及光訊號偵測系統固定不動。通常一個高靈敏度的表面電漿波生醫檢測裝置需要固定的光入射角,這是因為其需要複雜並且穩固的光學元件組。此固定入射角的特性卻限制了量測範圍,將同樣的檢測方式運用在此平台上將可獲得可調式光入射角的特性,如此就可獲得高靈敏度及大量測範圍的特性。具此,我們將可建造一高靈敏度平台並可運用在檢測物體或分子在不同的溶劑或背景液體中。研究結果證明其共振角檢測模式的靈敏度可達 0.00005 RIU,強度模式的靈敏度可達1x10^-5 RIU, 檢測範圍可由空氣到折射率大於水溶液。 A pair of off-axis parabolic mirrors was employed to construct a surface plasmon resonance (SPR) biosensing system as a universal platform for different interrogation methods. Utilization of parabolic mirrors ensures that this biosensor system is capable of an adjustable excitation angle in a large tuning range while the light source and interrogation system stay stationary. While a high-sensitive SPR biosensor is designed in a fixed-angle configuration because of its precise/complex optical alignment, the same scheme can be employed into this platform to obtain the tunable angle. An SPR biosensor with high sensitivity but in a much larger dynamic range can be obtained. High sensitivity test of molecular binding under different solvents is thus possible. The sensitivity test revealed that the sensitivity of angle interrogation is around 0.00005 RIU and that of intensity interrogation is around 1x10^-5 RIU. The dynamic range can cover solvents from air to water and beyond.
