生物可降解感溫水膠、以其作為載體之藥物遞送系統及用於治療和/或預防眼睛疾病的藥學組成物

本發明揭示一新式兼具降解與感溫性之注射型水膠作為眼部藥物傳輸系統,以克服藥物之低生物可利用率,並達到藥物緩釋的效果。此水膠材料乃由生物可降解之天然高分子明膠與感溫性聚異丙基丙烯醯胺所共聚而成,其最低臨界溶解溫度約在32°C。當材料注射至眼前房(約34℃),因環境溫度高於最低臨界溶解溫度,致始其迅速成膠並同時將大量藥物包覆至水膠內(包覆率約60%),改善傳統局部施加眼藥水須經許多屏障而難以到達眼內之缺點。而藉由明膠之降解,水膠載體可緩慢且持續地釋出藥物。以青光眼動物模式評估此傳輸系統,結果顯示藥物自水膠釋出後可持續降低眼壓效果達14天以上。 The present invention developed an injectable hydrogels for sustained intraocular drug delivery applications. A copolymer hydrogel was composed of thermally responsive and biodegradable polymers. In a particular aspect, the biodegradable polymer was gelatin, and the thermally responsive polymer was Poly(N-isopropylacrylamide) (PNIPAAm). Above its lower critical solution temperature (LCST) (i.e., around 32°C), the hydrogels featured a rapid sol-gel transition within seconds, which allowed in situ drug-incorporated gel formation. The biodegradation of gelatin was beneficial to form delivery systems with sustained drug release. In an animal model of glaucoma, the intraocular pressure (IOP)-lowering effect due to released drug was attained over at least 14 days. The copolymers were compatible with anterior segment tissues. Our findings suggest that the gelatin-g-PNIPAAm was an excellent candidate for use as a depot formulation for administration via intraocular injection.
