具雙影像追蹤探針之高分子奈米載體及其製備方法POLYMERIC NANOCARRIERS WITH DUAL IMAGES TRACKING PROBE AND METHOD FOR MANUFACTURING THE SAME

本發明開發雙影像追蹤之藥物奈米載體,即其奈米載體(nanocarriers)同時具有攜帶油溶性藥物與兩種影像追蹤等功能,即此奈米載體組成包含可發射螢光(fluorescence)與電腦斷層掃描(CT)影像追蹤系統的奈米金團簇(gold nanoclusters, AuNCs)。若應用PEG抗體接受奈米粒子表面PEG分子或接枝標靶配體(ligand)於奈米粒子表面,則使得此奈米載體具有標靶與雙影像追蹤的能力,可在給要同時去追蹤奈米粒子藥物載體,即大大提高即時判別藥物治療的效果的評估。方法如下:先將奈米金團簇(gold nanoclusters, AuNCs)接枝在聚乳酸-甘醇酸(poly(lactic-co-glycolic acid);PLGA)高分子末端成PLGA-AuNC,並另將PLGA接枝上聚乙二醇(polyethylene glycol;PEG)合成出PLGA-mPEG兩段式共聚高分子,再依兩者不同比例混合析出複合型奈米粒子。故本發明目標有1、合成與分析近紅外光之AuNCs。2、合成與鑑定PLGA接枝AuNCs。3、合成與鑑定PLGA接枝PEG。4、依PLGA-AuNC與PLGA-mPEG兩者不同比例混合析出複合型奈米粒子,並評估其奈米粒子特性與包覆攜帶藥物的能力。5、探討此多功能性之腫瘤標靶奈米載體之影像追蹤能力,並結合anti-PEG抗體對PEG之專一性標靶能力評估。本發明已成功合成出高生物相容性與多功能性之腫瘤標靶奈米載體,即其同時具有攜帶油溶性藥物、PEG抗體腫瘤標定與奈米載體的近紅外光、CT影像追蹤等功能。 This study intends to develop versatile of the tumorous targeting nanocarriers that also have to carry cancer drugs, tumorous targeting and dual system images and so on. This tumorous targeting nanocarriers composed of nanogold clusters (AuNCs) grafted on PLGA into the end of the copolymer PLGA (PLGA-AuNC). Besides, PLGA was also grafted with PEG (polyethylene glycol) to PLGA-mPEG copolymer. The final complex nanoparticles product was mixed in different proportions of PLGA-AuNC and PLGA-mPEG. Therefore, there are objectives of the this experiment. (1) Synthesis and analysis near-infrared of AuNCs. (2) Synthesis and characterization of PLGA-AuNCs. (3) Synthesis and characterization of PLGAPEG. (4) The PLGA-AuNC mixed with PLGA-mPEG in different proportions to complex nanoparticles and evaluated its characteristics. (5) To evaluate this nanocarriers for tracking compatibility of tumorous targeting image and identify the target capacity of PEG-PLGA nanocarriers by anti-PEG antibodies.
