用於抑制組蛋白基因轉錄及表現之醫藥組合物及其應用

扁柏油酚(hinokitiol),可由柏科植物萃取,其抗菌功效已廣為人知,也被應用在化妝品、食品添加劑中,但抗癌功效目前僅止於研究,尚無任何臨床應用。本人實驗發現扁柏油酚可抑制口腔蛀牙與牙周病細菌孳生。口腔癌細胞毒性具專一性,對正常細胞毒性較弱,甚至減少腫瘤幹細胞的標誌蛋白。實驗證據顯示扁柏油酚可能藉由抑制組蛋白轉錄,使細胞周期停止在S期,目前臨床抗癌用藥無法同時兼具抗菌功能,或藥物副作用極強。扁柏油酚同時兼具抗菌及抗癌特性,抗癌機轉特殊,極具開發潛能。 Hinokitiol is a constituent of the woody oils isolated from Cupressaceae Heartwood. It has been found to be act as antibacterial agent, cosmetics addition, and natural food preservative. However, hinokitiol still be study in anti-cancer field. We found that hinokitiol can inhibit caries pathogen, Streptococcus mutans, and periodontal disease pathogen, Actinobacillus actinomycetemcomitans. The cytotoxicity of hinokitiol was higher in oral squmaous cell carcinoma (OSCC) cell lines than normal oral cells. Even cancer stem cells marker decrease after hinokitiol treatment. Hinokitiol treated with OSCC cell lines, and the cycle arrest in S phase by histone gene transcript inhibition. There are few clinical chemotherapy drugs with both anti-microorganisms and anti-cancer capability at the same time. Most anticancer drugs have distinct side effects. Hinokitiol has anti-microorganism and anti-cancer bifunction, and the anti-cancer mechanism is different from other drugs.
