二茂鐵化合物及用於檢測沙門氏菌的樣品分析方法

沙門氏菌的污染是食品和公共衛生安全的主要問題,檢測食品中的沙門氏桿菌是保障民眾健康和產業經濟的重要步驟。因此需要開發一個易於操作的沙門氏菌檢測平台。為此我們設計了兩種活性基礎的潛伏式比率電化學探針Sal-CAF 和Sal-NBAF,專門用於通過簡單的稀釋實現對沙門氏菌酯酶(一種沙門氏菌生物標誌物)的簡單、快速和選擇性的分析。探針的辛酯被辛酯酶水解並觸發三甲基鎖以彈出含電子的氨基二茂鐵衍生物,並使用電化學方法檢測到相應的訊號。Sal-CAF探針擁有大偵測範圍以及低檢測極限定量活沙門氏菌的濃度,並成功地以最少的樣品前處理定量牛奶樣品中摻入的活沙門氏菌。 Salmonella contamination is a major concern in food and public health safety; episodic monitoring of Salmonella contamination in food is essential to safeguard public health and economy. Therefore, there is a need to develop an easy-to-operate Salmonella-targeting point-of-care detection platform. To this, we designed two activity-based latent ratiometric electrochemical molecular probes, Sal-CAF and Sal-NBAF, specifically for achieving easy, rapid, and selective profiling of Salmonella esterase (a Salmonella biomarker), with simple dilution. The octyl esters of the probe were cleaved by the C8 esterase and trigger the trimethyl lock to eject the electron-rich aminoferrocene derivatives, and the corresponding electrochemical signals were detected by electrochemical method. The Sal-CAF probe was used to determine the concentration of live Salmonella in a wide detection range with a low detection limit. The developed Sal-CAF probe was successful in quantifing live Salmonella spiked in milk samples with minimal sample pretreatments
