APPLICATION OF AVOCADO EXTRACT, AVOCADENOL B, AND (2R,4R)-1,2,4-TRIHYDROXY HEPTADEC-16-YNE, AND HEALTH FOOD COMPRISING AVOCADO EXTRACT

1-1. 樣本採集: 未成熟之酪梨果實採集於夏天,採集地點為高雄地區,採集之樣本先行物種鑑定,接著以清水清洗表面,並且將其果肉與種子分離後,將果肉切成薄片,保存在零下40度的環境中。 1-2. 萃取及存化: 未成熟之酪梨果實切片後,放在50℃之恆溫烘箱內烘乾。乾燥後的樣本首先利用甲醇(MeOH)放於室溫中萃取三次。直到甲醇萃取物內之EtOAc–H2O純化比例為1:1則將兩者分成EtOAc-soluble fraction與H2O-soluble fraction。其中EtOAc-soluble fraction則為本次實驗具有生物活性之酪梨甲醇萃取物(簡稱:MEPA)。隨後將MEPA以矽膠(silica gel)分離,利用濃度梯度之n-hexane–EtOAc洗滌後得到16個分層(A-1到A-12),其中A-12為有生物活性之分層。隨後我們將A-12層利用n-hexane重新結晶化得到crystals fraction(A-12-C)以及mother liquor fraction(A-12-m)。緊接著將A-12-m以silica gel分離並且利用濃度梯度之hexane–acetone洗滌後得到七個分層(A-12-m-1到A-12-m-7)。A-12-m-4則利用RP-C18 column進行色層分析,並且利用acetone–H2O (1:1)進行洗滌,最後得到Avocadenol B 以及(2R,4R)-1,2,4-trihydroxyheptadec-16-yne ,此兩小分子化合物即為本計劃中具有生物活性之酪梨萃取小分子純化合物。 P. americana were sliced , dried and stored at -40℃. The unripe fruits of P. americana were sliced and dried by constant temperature oven (50℃) to obtain the dried unripe pulp. The dried unripe pulp was extracted with MeOH at room temperature three times. Upon concentration, the MeOH extract was partitioned between EtOAc–H2O (1:1) to obtain an EtOAc-soluble fraction and an H2O-soluble fraction. An active EtOAc-soluble fraction was applied to a silica gel column, eluting with a gradient of n-hexane–EtOAc, to give 16 fractions (A-1–A-16). Fraction A-12 was recrystallized from n-hexane to give crystals (A-12-c) and the mother liquor (A-12-m). Fraction A-12-m was submitted to a column containing silica gel, eluting with a gradient of n-hexane–acetone, to yield seven fractions (A-12-m-1–A-12-m-7). Fraction A-12-m-4 was chromatographed on a RP-C18 column to obtain bioactive constitutes including Avocadenol B and (2R,4R)-1,2,4-trihydroxyheptadec-16-yne.
