一種用以生成蘭花植物多倍體植株的方法method for producing polyploid plants of Orchids

本發明之特徵在於提供一種不用特殊化學藥劑而以植物組織培養技術將植體中含有細胞內生多倍化的組織培育成多倍體植株的方法。本發明的方法包括在無菌操作環境下,利用植物組織培養及切割蝴蝶蘭不同組織的技術,在培養基中誘導擬原球體(PLB)的發生。當新誘導的PLB長至合適大小,可進行切割步驟以增加PLB的數量及多倍體的發生率。其後,以適當培養基將PLB培育成具葉及根的小植株;再以流式細胞分析儀進行染色體倍數檢定。本發明的方法簡單、實用而有效,能產生大量穩定的蝴蝶蘭多倍體植株,包括其品種及雜交種。 Disclosed is a method for production of polyploids of Phalaenopsis orchids without using special chemical agents, but through plant tissue culture to regenerate polyploidy plants from tissues having endopolyploid cells. The method includes cutting of suitable plant tissues under sterile condition to produce protocorm-like bodies (PLB) using PLB induction medium. Repeat the above cutting procedure when the newly developed PLB reached the appropriate size in order to increase the number of PLB and the frequency of ployploids. Then the induced PLB re-differentiate into plantlets with leaves and roots in a re-differentiation medium. Then the ploidy level of the plant is determined by using flow-cytometer. The technique is simple, practicable and effective in producing stable polyploids of Phalaenopsis including varieties and related hybrids.
