胜肽之胺基酸序列的測定方法與系統

本發明揭露胜肽之胺基酸序列的測定系統與方法。首先,將待測蛋白質水解成短胜肽與胺基酸。接著,以高效液相層析分離短胜肽與胺基酸、定性辨識除半胱胺酸外之胺基酸種類,以及利用一質譜儀測定短胜肽的分子量與半胱胺酸。接著,以由小至大的順序,經由測得的胺基酸組合構成各種短胜肽,比對質譜之分子量訊號,確認該短胜肽之胺基酸序列,繼續組合確認的短胜肽構成較大之各種胜肽,再比對質譜之分子量訊號,確認較大之胜肽的胺基酸序列,依此程序直到完成整個蛋白質胜肽鍊之胺基酸序列。 This invention discloses systems and methods for determining the sequence of amino acids in a short peptide chain that constructs a protein. The protein is firstly hydrolyzed to a variety of short peptides and amino acids. Then, the high-performance liquid chromatography (HPLC) systems and method are used to separate the short peptides and the amino acids, identify qualitatively each of the amino acids except cysteine, and obtain the molecular mass for each of the peptides and identify cysteine by a mass-spectrometry measurement. After that, the identified amino acids are used to construct any possible short peptides in an order from the smallest molecular weight dipeptide to higher molecular weight short peptides, and the correct short peptides is confirmed by matching the molecular weight obtained from the mass-spectrometry measurement, then, the short peptides are combined to give a large peptide.
