技術說明

本發明揭露由海綿動物萃取出的抗癌manoalide化合物、其醫藥組成物、製備方法及構型鑑定方法。該化合物具有式I結構式，其中，R為CH3，C-24及C-25的構型組合為24R,25R、24R,25S或24S,25S。式I化合物是以乙酸乙酯萃取海綿動物、濃縮乙酸乙酯萃取物獲得殘留物、層析該殘留物獲得多個分餾物、以及通過矽膠管柱層析以及反相高效液相層析分離而獲得。將式I化合物與標準品的質子化學偏移進行比對，以判斷式I化合物的構型。 Disclosed are antiproliferative manoalide compounds extracted from the sponge Luffariella sp., their pharamaceutical composition, preparation method and identification method for the configuration. The compound has a structural formula (I), where R is CH3, the configuration combination for C-24 and C-25 is 24R,25R, 24R,25S or 24S,25S. The compound (I) is obtained by extracting Luffariella sp. with ethyl acetate (EtOAc), concentrating the EtOAc extract to obtain a residue, chromatographing the residue to yield fractions, and separating the compound (I) from fractions by the column chromatography on silica gel and reverse-phase HPLC. The proton chemical shifts of the compound (I) are compared with those of the standards to determine the configurations of the compound (I).
