技術說明

本發明主要是揭示一種Cell Sheet Engineering系統；藉由不同包括Tissue Culture Polystyrene、Polyethylene及Polyvinyl difluoride、玻璃等透明性的底材，包括緻密性及多孔質的表面來作為單層及複層片狀細胞層的基材，並在基材上進行具雙硫鍵細胞親合性胺基酸分子的表面電漿處理並進行天然高分子與之交聯，再以數種細胞進行細胞培養，並觀察細胞在滿盤之後以還原性胺基酸處理使之成為Cell Sheet；期望能以Cell sheet的概念，讓培養成層狀的細胞能在完整保留細胞外間質的狀態下脫離培養的表面，以提高培養細胞養分供應的效率，可進一步應用在人類自體組織培養及移植上。 It is intended to provide a cultured cell sheet with excellent tissue adherence and flexibility. The above object can be achieved by culturing cells on a support for cell culture in which a surface of a substrate is coated with a natural biopolymer via a disulfur bond-containing amino acid as a spacer. To cut the disulfur bonding between cultured cell sheet and culture matrix and producing cell sheet by detaching it by a reducing amino acid. Look forward to the concept of cell sheet engineering, so that cells can develop into a layer of extracellular matrix in the remains intact state from the culture surface to improve the efficiency of nutrient supply to cultured cells can apply the further applications of human autografting related products.
