用於誘導細胞凋亡之樟芝子實體乙醇萃取物及其製備方法Ethanol extract of Antrodia camphorata for inducing apoptosis and preparation method thereof

本專利係提供一種樟芝子實體乙醇萃取物的製備方法,包括下列步驟:(a)提供一樟芝子實體;及(b)以一乙醇溶液萃取該樟芝子實體,過濾並離心,獲得該樟芝子實體乙醇萃取物。該樟芝子實體乙醇萃取物還可以正己烷、乙酸乙酯及乙醇依序萃取或分餾,分別獲得樟芝子實體正己烷(FC)、乙酸乙酯萃取物(FA)及乙醇再萃取物(FB)。乙酸乙酯萃取物(FA)可有效抑制血癌細胞生長並促進細胞凋亡,其內含活性成分—樟芝酸A,可做為生物活性分子。並提供一檢測樟芝子實體三萜類含量之方法,檢測方法包括核磁共振圖譜與高效液相層析法。 A preparation method for an ethanol extract of the fruiting body of Antrodia camphorata (EEAC) is provided. The preparation method includes steps of: (a) providing the fruiting body of A. camphorata (AC); (b) extracting the fruiting bodies with a first ethanol solution; and (c) obtaining EEAC. EEAC further can be sequentially extracted or fractioned by n-hexane, ethyl acetate and ethanol, and an n-hexane fraction (FC), an ethyl acetate fraction (FA) and an ethanol fraction (FB) respectively are generated. The growth inhibition and apoptosis induction of leukemia cell line HL 60 are effectively mediated by FA product, in which zhankuic acid A is the bioactive marker. The amount of triterpenoid in the fruiting body of AC can be determined by NMR and HPLC analysis.
