技術說明

薊馬是植物花朵、蔬菜或果樹上常見的害蟲,甚至會傳染植物病毒;薊馬幼蟲及成蟲取食植物組織,造成葉芽、花部及果實等扭曲、變矮、及斑紋等損害,影響作物之品質及產量。其體形細小,借由玻片製作及顯微鏡以傳統形態鑑定方式鑑定成蟲,但薊馬幼蟲則仍無法辨別。本研究以農業上危害嚴重的薊馬為材料,收集欲鑑別之薊馬樣本8種,萃取薊馬的去氧核醣核酸,進行定序並分析各種薊馬的序列,以各種薊馬之核醣體基因間隔2區DNA序列設計專一性引子,經過專一性及穩定性測試,應用於近20種薊馬之鑑定。本發明提供鑑定八種重要薊馬害蟲之引子對,及利用上述引子對鑑定薊馬害蟲之方法。 Thrips are major pests of many agricultural plants worldwide, including staple crops, floral plants, vegetables and fruit trees. As the only vectors of plant Tospovirus, they can directly damage the host plants by attacking twigs, leaves, buds, flowers, and fruits resulting in stunted growth, feeding scars, distortions, and color mosaic. The identification of thrips has been traditionally based on morphological characters of adult specimens, visible via slide mounting and through microscopic examination. However, their minute size and insufficient phenotypic variation, makes adults difficult to identify and larvae generally impossible to identify to species level. In this study, species-specific primers were designed based on the constructed ITS2 sequences of 8 agriculturally important thrips). The efficacy and specificity of these primers were examined, respectively, on conspecific individuals and a total of ca. 20 thrips species.
