用於治療糖尿病及新陳代謝的組合物及其製備方法Composition for treating diabetes and metabolic diseases and a preparation method thereof

查耳酮類(Chalcones)為天然物中常見之黃酮類骨架化合物。文獻上已經有相當多的活性甚至機轉之研究,但至今仍鮮少有關抗糖尿病的研究。本團隊以脂肪細胞、胰臟beta 細胞、及肝臟細胞為測試平台,篩選化學資料庫之化合物,觀察其是否能增加葡萄糖利用率、改善胰島素敏感性、促進胰臟beta 細胞分泌胰島素、及促進肝臟細胞肝醣合成。結果發現黃酮類中查耳酮骨架(Chalcones)之AN07 系列化合物為此模式投與藥物中最有效之化合物骨架,更進一步以高脂肪餵食誘發胰島素抗阻性糖尿病小鼠,評估AN07 最佳衍生物對降低血糖的效果。本發明係提供一種具有抗糖尿病效果的組合物及其合成方法,其中本發明利用化學方法合成多種新穎的黃酮類化合物,利用不同的起始物、反應物及中間產物,產生多種不同之衍生物。 Chalcone is a kind of well-known flavonoids. So far, there are a lot of active and mechanical evidences on it but rare anti-diabetes mellitus related researches. We use adipocyte, hepatocyte, and insulin producing cell lines to evaluate whether compounds from our chemical bank could improve glucose utilization, insulin sensitivity, glycogen production, insulin production, and beta cell protection. The results showed that AN07 series compounds (chalcone derivates) were the most potential candidates. The high-fat diet feeding combine with the streptozotocin-induced diabetic mice model was used to evaluate compound’s efficacy on anti-hyperglycemic ability. The present invention provides a composition having the anti-diabetic effect and a synthesis method for preparation flavonoids, wherein the novel flavonoids include derivatives that synthesized from different initiators, reactants, and intermediates.
