單細胞擷取與培養之裝置與方法

胚胎幹細胞保有發展成體內各種類型細胞的潛能。而類胚胎體的形成在胚胎幹細胞分化過程中扮演了重要的角色。傳統上最廣泛使用來形成胚胎體的方式為”懸吊式液滴技術”。此方式通常是把胚胎幹細胞懸浮液點在細胞培養皿的上蓋,形成液珠的形狀,因著地心引力的關係使胚胎幹細胞聚集在液滴的底部。然而,這個方法需要花費人力和時間,在更換培養液上面也比較困難。為了解決這些問題,本計畫欲發展了一組微流體陣列式的懸吊式液珠晶片,可以高通量產生液滴。我們將利用微流道技術同時形成大量之個別液滴,並且可以不需對個別液滴進行細胞載入動作,進而降低操作時間。晶片當中的微流道也會使液滴培養液置換變得容易,提供長時間細胞培養之可行性。另外,微小化之液滴可以節省昂貴之培養液以及試劑對,可以讓此技術衍生的產品更具市場競爭力 Embryonic stem cells have the potential to develop into all types of cells in animal tissues. Embryoid body (EB) formation is an import step in embryonic stem cell culture and differentiation. Conventional hanging drop technique widely used for EB formation, is based on hanging ES cell suspension as small droplets on the lid of petri dish to let ES cells to gather at the bottom of the droplet by gravity. However, this method is labor intensive and limited by the difficulty in exchanging medium. To overcome the above limitations, we aim to develop an arrayed micro-hanging drop platform which can simultaneously form large numbers of hanging drops for EB formation. Our device will also allows for medium exchange making long-term culture of EBs possible in our hanging drop device. In addition, the consumptions of medium and reagents can be reduced by using miniaturized drops in our device. This could significantly make our aim product more competitive to existing products in the market.
