技術說明

本發明提出螢光奈米鑽石的製造方法與生物醫學方面的應用。我們首先以一離子束轟擊奈米鑽石,產生晶格缺陷後,在高溫下加熱退火完成。實驗證明當這些鑽石微晶粒受到藍綠光照射時,其氮原子-空隙所形成的缺陷會發出強烈的綠色或紅色螢光。以此方法製成之螢光奈米鑽石的品質高,光穩定性極佳,完全不會有光漂白及光閃爍的現象,可以增長觀察的時間,單一顆螢光奈米鑽石可以很容易的以共軛焦螢光顯微鏡觀察到。螢光奈米鑽石不僅僅有持久螢光、可耐極端化學條件的特性,其生物毒性也非常的低,對於細胞體正常生理幾乎不會造成影響,因此可作生物細胞造影與長期追蹤之用。同時,奈米鑽石的紅色螢光對生物組織有相當好的穿透力,可應用於活體的生物顯影,這項優點使得螢光奈米鑽石更有開發與研究的價值。另外值得一提的是,奈米鑽石晶粒表面經過氧化酸處理後,很容易攜帶有含氧的官能基,此官能基可以進一步以化學方法來修飾而有特殊的性質。透過這些新的表面特性,表面修飾過的奈米鑽石可以當作基因轉植的載體,假若再接上如抗原般具有特異性作用力的生物分子後,奈米鑽石在生物系統中就具備有辨識目標的能力,是一項長期應用於活體生理現象追蹤、顯影的新工具。 We disclose a new method to produce bright fluorescence nanodiamonds (FNDs) economically and in a large scale for biomedical applications. It involves creation of high concentrations of color centers using a low-mass ion beam (such as H+ and He+). In particular, a method for mass production of FNDs with a commercially available, medium-energy, high-fluence ion source is demonstrated. Additionally, we illustrate novel applications of this material in life sciences using both confocal and epifluorescence microscopies. Observations of single FND particles deposited on a glass substrate and in a live cell are also presented. The optical properties, such as photobleaching and fluorescence intermittency (blinking), are closely characterized for individual FNDs. It is found that FNDs are exceptionally photostable under high-power laser excitation (up to an intensity of 1 MW/cm2) and the brightness of the fluorescence remains almost at the same level even after covalent immobilization of biomolecules such as polypeptides and DNA onto their surfaces. This invention demonstrates that FND is a promising nanomaterial to be used as a fluorescent nanoprobe for in vitro as well as in vivo studies at the single particle level.
