技術說明

骨質疏鬆是高齡化社會重要保健課題。目前雙磷酸鹽是治療骨質疏鬆最常用的藥物，磷酸鹽一般的副作用是在胃腸系統，一些資料顯示長期服用雙磷酸鹽，雖然很少，但是會發生很嚴重下顎骨壞死的情形。一般認為草藥的副作用少，因此較適合於長期服用的情形。傳統的台灣草藥台灣骨碎補通常用於強骨，且宣稱有促使骨骼痊癒的效果。我們先前的研究已確認台灣骨碎補的安全性及抗骨質疏鬆的效果。這些結果足以支持台灣骨碎補開發成抗骨質疏鬆的健康食品。未來的研究將與食品業廠商合作。 在第一年，我們將確認台灣骨碎補配方對去卵巢大鼠抗骨質疏鬆的效果，我門也將評估台灣骨碎補配方的急性毒性、28天餵食毒性。以生物活性導向的原則，分離抑制具噬細胞RAW264.7分化成破骨細胞的成分。在第二年，將評估台灣骨碎補配方的90天餵食毒性及致畸胎毒性。我們也將探討有效成分在細胞的作用機轉。還有，將建立有效成分HPLC分析系統用來控管產品。由於有效成分無法在市場上買到，因此大量製備提供廠商是必要的。 集合這些數據，台灣骨碎補配方將提出抗骨質疏鬆健康食品之申請。 In the first year, we will confirm the benefit of formulas of D. formosana on osteoporosis in ovariectomized rats. We will evaluate the acute toxicity, 28-day feeding toxicity for formulas of D. formosana. On the basis of the bioactivity-guided fractionation principle, we will isolate the active components for inhibiting the formations of osteoclast from RAW 264.7 cells. In the second year, the evaluation of 90-day feeding toxicity, teratogenicity, genotoxicity for formulas of D. formosana. will perform. We will also examine the mechanisms for active component on in vitro studies. Furthermore, the analytic system used the HPLC for active compound will establish for the quality control of product. Since this active compound can not purchase from market, it is necessary that we will large scale purification of this compound provide the cooperated manufactory. Taken together these data, formula of D. formosana will apply for health food for the preventing osteoporosis
